چکیده

33 The aim of this work was to study the effects of mastitis 34 induced by intramammary lipopolysaccharide challenge on milk 35 oxidative stability, as well as to understand the underlying 36 biochemical processes that cause such changes. 37 Lipopolysaccharide challenge was associated with nitric oxide 38 burst from surrounding mammary epithelial cells and 39 consequently provoked nitrosative stress that was induced by 40 formation of NO2• from nitrite by lactoperoxidase. This response 41 was associated with ~3 fold increased formation of hazardous 42 compounds: nitrotyrosines, carbonyls and lipid peroxides. We 43 sustained the involvement of xanthine oxidase as a major source 44 of hydrogen peroxide. In consistent with previous findings, 45 catalase has been shown to have a major role in modulating the 46 nitrosative stress by oxidizing nitrite, to nitrate. The current 47 hygienic quality criteria cannot detect mixing of low-quality milk, 48 such as milk with high somatic cells and nitrite with high-quality 49 milk. Thus, developing of improved quality control methodology 50 may be found important for the production of high quality milk. 51 4 52

Changes of stress oxidative enzymes in rat mammary tissue, blood and milk after experimental mastitis induced by E. coli lipopolysaccharide

The present study investigated the effects of E. coli lipopolysaccharide (LPS) induced mastitis model in rat on the activity of antioxidant enzyme systems. To achieve this purpose, E. coli LPS were infused into the mammary gland of 12 rats 72 hr postpartum and compared with 12 rats in control group infused intramammary placebo sterile pyrogene – free, physiological saline. The...

Lipopolysaccharide challenge of the mammary gland in bovine induced a transient glandular shift to anaerobic metabolism.

Support of milk production in modern dairy cows demands a large proportion of its own metabolic resources, such as glucose, which might be required under stressful situations. The aim of the experiment was to test the hypothesis that acute immune stress shifts oxidative metabolism to glycolysis. Two mammary quarters in 6 Holstein cows were infused with lipopolysaccharide (LPS), whereas the 2 co...

Effects of heat stress and dietary zinc source on performance and mammary epithelial integrity of lactating dairy cows.

Dietary Zn and heat stress alter gut integrity in monogastric animals. However, effects of Zn on mammary epithelial integrity in heat-stressed lactating dairy cows have not been studied. Multiparous lactating Holstein cows (n = 72) were randomly assigned to 1 of 4 treatments with a 2 × 2 factorial arrangement to study the effects of environment and Zn source on performance and mammary epithelia...

Mammary gene expression and activity of antioxidant enzymes and oxidative indicators in the blood, milk, mammary tissue and ruminal fluid of dairy cows fed flax meal.

The effects of flax meal (FM) on the activity of antioxidant enzymes (superoxide dismutase (SOD), glutathione peroxidase (GPx) and catalase (CAT)) in the blood, mammary tissue and ruminal fluid, and oxidative stress indicators (thiobarbituric acid-reactive substances(TBARS) and 1,1-diphenyl-2-picrylhydrazyl-scavenging activity) in the milk, plasma and ruminal fluid of dairy cows were determined...

Increase of Escherichia coli inoculum doses induces faster innate immune response in primiparous cows.

The objective of the current study was to evaluate the dynamics of infection and the immunological response to varying numbers of Escherichia coli injected into the mammary glands of primiparous cows during the periparturient period. Primiparous cows have been shown to be more resistant to intramammary E. coli challenge, and an increase of the inoculum dose by 2 log10 units induced a more rapid...
